    Diving in Two Marine Lakes in Croatia

    We describe the diving methods used for in-situ observations of the scyphozoan medusa, Aurelia sp., in two marine lakes on the island of Mljet, Croatia. Both lakes have a strong pycnocline at approximately 15 m. During this study (May, 2004) surface temperature was about 20o C; bottom temperature about 10o C. Visibility was 15 m to 30 m. Tide and currents were negligible. A dense resident population of Aurelia sp. and a predictable environment made this an ideal study site. Aurelia was most abundant in mid-water around the pycnocline. There were several dive objectives: specimen collection for laboratory analysis, population census, discrete plankton tows and direct observation of flow around swimming medusae. We used several methods for maintaining our orientation underwater including working from an anchor line, towing a tethered buoy, and use of a blue water rig. Because the environment was relatively benign we allowed the rig to drift free while the boat was standing by at a short distance. Often a tether was not required. This plan allowed the most freedom and provided an excellent reference throughout the dive

    Omnivory by the Small Cosmopolitan Hydromedusa Aglaura Hemistoma

    We investigated the feeding of the small hydromedusa, Aglaura hemistoma (bell diameter \u3c 4 mm), to determine if it occupies a trophic position similar to that of large medusae. Feeding was examined using gut-content analysis of preserved and unpreserved medusae and by analyzing prey-capture events using microvideographic techniques. Analysis of gut contents and prey-capture events revealed that A. hemistoma fed heavily on protistan prey and that it possessed a prey-capture mechanism, specifically a feeding current,that is effective at entraining and capturing protists with low motility. We suggest that many species of small hydromedusae possess prey-capture mechanisms adapted to capture small protistan prey and that many of these small hydromedusae feed omnivorously on microplanktonic prey. The trophic roles of small hydromedusae in different systems are not understood and more studies are needed. However, based on their often high abundances and the cosmopolitan nature, if small hydromedusae are primarily omnivores, they need to be considered when estimating the impact of zooplankton on primary production and, more generally, protistan community dynamics

    Absolute photoionization cross section measurements of the Kr I-isoelectronic sequence

    Photoionization spectra have been recorded in the 4s, 4p and 3d resonance regions for the Kr Iisoelectronic sequence using both the dual laser produced plasma technique (at DCU) to produce photoabsorption spectra, and the merged ion beam and synchrotron radiation technique (at ASTRID) to measure absolute photoionization cross sections. Profile parameters are compared for the 4s − np resonances of Rb+ and Sr2+. Many new 4p " ns, md transitions are identified with the aid of Hartree-Fock calculations, and consistent quantum defects are observed for the various ns and md Rydberg series. Absolute single and double photoionization cross sections recorded in the 3d region for Rb+ and Sr2+ ions show preferential decay via double photoionization. This is only the second report where both the DLP technique and the merged beam technique have been used simultaneously to record photoionization spectra, and the advantages of both techniques (i.e. better resolution in the case of DLP and values for absolute photoionization cross sections in the case of the merged beam technique) are highlighted

    The interchangeability, confusion and conflict of what constitutes audience engagement has a long history, with much disagreement concerning boundaries and definitions. Dewey states that it is a mistake to see the artist as active and the audience as purely passive, and argues that “the active engagement of the audience is required to fully realise any work” (Dewey 1934). This predates the notions of “interactive” or “participatory” as understood today, but highlights the longstanding appreciation of the role the audience plays in the consumption of artworks. A sentiment echoed by Duchamp (1957) stating that “the spectator adds his contribution to the creative act”. The research project presented at EVA 2017 seeks to offer a model for engagement, that of pleasure, which explores methods to motivate active participation

    Gene transfer into stimulated and unstimulated T lymphocytes by HIV-1-derived lentiviral vectors

    Genetic modification of T lymphocytes holds great potential for treatments of cancer, T cell disorders and AIDS. While in the past recombinant murine retroviruses were the vectors of choice for gene delivery to T cells, vectors based on lentiviruses can provide additional benefits. Here, we show that VSV-G pseudotyped HIV 1 vector particles delivering the enhanced green fluorescent protein (EGFP) efficiently transduce human T lymphocytes. Transduction efficiency was optimal when infection included centrifugation of cells with concentrated vector supernatant in the presence of Polybrene. In contrast to previous reports describing murine retrovirus-mediated gene transfer to T lymphocytes, fibronectin did not improve the transduction efficiency of the VSVG-pseudotyped HIV-1 particles. Similar gene transfer efficiencies were observed following stimulation of cells with PHA/IL-2 or anti-CD3i/CD28i antibodies, although greater transgene expression was observed in the latter case. Interestingly, production of vectors in the absence of the accessory proteins Vif, Vpr, Vpu and Nef was accompanied by a 50% decrease in transduction efficiency in activated T cells. Transduction of T cells that were not stimulated before infection was achieved. No transduction of non-prestimulated cells was observed with a GAL V-pseudotyped murine retroviral vector. The requirement for accessory proteins in non-prestimulated cells was more pronounced. Our results have implications for lentiviral vector targeting of other cells of the hematopoietic system including stem cells

    A Census of Marine Biodiversity Knowledge, Resources, and Future Challenges.

    ABSTRACT. The Census of Marine Life (2000–2010) was the largest global research programme on marine biodiversity. This paper integrated the findings of reviews of major world regions by the Census and provides a global perspective on what is known and what are the major scientific gaps. Study metrics were regional species richness, numbers of endemic and alien species, numbers of species identification guides and taxonomic experts, and a state-of-knowledge index. The threats to biodiversity were classified across the regions. A poor to moderate correlation between species richness and seabed area, and sea volume, and no correlations with topographic variation, were attributed to sparse, uneven and unrepresentative sampling in much of the global marine environment. Many habitats have been poorly sampled, particularly in deeper seas, and several species-rich taxonomic groups, especially of smaller organisms, remain poorly studied. Crustacea, Mollusca, and Pisces comprised approximately half of all known species across the regions. The proportion that these and other taxa comprised of all taxa varied sufficiently to question whether the relative number of species within phyla and classes are constant throughout the world. Overfishing and pollution were identified as the main threats to biodiversity across all regions, followed by alien species, altered temperature, acidification, and hypoxia, although their relative importance varied among regions. The findings were replicated worldwide, in both developed and developing countries: i.e. major gaps exist in sampling effort and taxonomic expertise that impair society's ability to discover new species and identify and understand species of economic and ecological importance. There was a positive relationship between the availability of species identification guides and knowledge of biodiversity, including the number of species and alien species. Available taxonomic guides and experts correlated negatively with endemic species, suggesting that the more we study the ocean the fewer endemic species are evident. There is a need to accelerate the discovery of marine biodiversity, since much of it may be lost without even being known. We discuss how international collaboration between developed and developing countries is essential for improving productivity in the discovery and management of marine biodiversity, and how various sectors may contribute to this

    Equivalences between GIT quotients of Landau-Ginzburg B-models

    We define the category of B-branes in a (not necessarily affine) Landau-Ginzburg B-model, incorporating the notion of R-charge. Our definition is a direct generalization of the category of perfect complexes. We then consider pairs of Landau-Ginzburg B-models that arise as different GIT quotients of a vector space by a one-dimensional torus, and show that for each such pair the two categories of B-branes are quasi-equivalent. In fact we produce a whole set of quasi-equivalences indexed by the integers, and show that the resulting auto-equivalences are all spherical twists.Comment: v3: Added two references.
